چکیده

As part of an investigation of a plant virus which multiplies within the cells of its aphid vector (O'Loughlin and Chambers, 1967), the fine structure of the aphid gut was studied. The alimentary canal of aphids commences with the food canal which is formed by the apposition of grooves in the two interlocking feeding stylets . The stylet food canal leads to the sucking or pharyngeal pump within the aphid head, and this carries food into the foregut . The foregut is a narrow tube which opens via the esophageal valve into the first part of the midgut, usually referred to as the "stomach." The second part of the midgut, the "intestine," follows a devious course and is the longest part of the alimentary canal . It finally widens to become the hindgut which is thin walled and highly distensible . Specimens of Hyperomyzus lactucae (L .), reared on sowthistle (Sonchus oleraceus L .), were cut in half transversely in cold 3 % glutaraldehyde and fixed for up to 14 hr, followed by postfixation in cold 1 0/o osmium tetroxide in sodium cacodylate buffer pH 7 .4. They were dehydrated in acetone and embedded in Araldite. Thin sections were stained with uranyl acetate followed by lead citrate . The striated border of the intestine of the aphid Myzus persicae (Sulzer) has been reported to consist of typical microvilli (Forbes, 1964), but our observations, initially on H. lactucae, show that one can resolve the area surrounding each microvillus as made up of numerous extracellular microtubules that lie close to and parallel with the BRIEF NOTES

Automated Solution-Phase Synthesis of Insect Glycans to Probe the Binding Affinity of Pea Enation Mosaic Virus

Pea enation mosaic virus (PEMV)--a plant RNA virus transmitted exclusively by aphids--causes disease in multiple food crops. However, the aphid-virus interactions required for disease transmission are poorly understood. For virus transmission, PEMV binds to a heavily glycosylated receptor aminopeptidase N in the pea aphid gut and is transcytosed across the gut epithelium into the aphid body cav...

Modification of Cry4Aa toward Improved Toxin Processing in the Gut of the Pea Aphid, Acyrthosiphon pisum

Aphids are sap-sucking insects (order: Hemiptera) that cause extensive damage to a wide range of agricultural crops. Our goal was to optimize a naturally occurring insecticidal crystalline (Cry) toxins produced by the soil-dwelling bacterium Bacillus thuringiensis for use against the pea aphid, Acyrthosiphon pisum. On the basis that activation of the Cry4Aa toxin is a rate-limiting factor contr...

Short Communication Spider Predation: Species-specific Identification of Gut Contents by Polymerase Chain Reaction

We extend detection of arthropod predator gut contents by polymerase chain reaction (PCR), heretofore restricted to insect predators, to spiders. Single individuals of the corn lead aphid, Rhopalosiphum maidis, were detected in the guts of spiderlings of Oxyopes salticus up to 12 h after feeding; individuals of the congeneric bird cherry oat aphid, R. padi, were not detected. Unfed O. salticus ...

Influence of taxol and CNTs on the stability analysis of protein microtubules

Microtubules are used as targets for anticancer drugs due to their crucial role in the process of mitosis. Recent studies show that carbon nanotubes (CNTs) can be classified as microtubule-stabilizing agents as they interact with protein microtubules (MTs), leading to interference in the mitosis process. CNTs hold a substantial promising application in cancer therapy in conjunction with other c...

The passage of Potato leafroll virus through Myzus persicae gut membrane regulates transmission efficiency.

Potato leafroll virus (PLRV) is transmitted by aphids in a persistent manner. Although virus circulation within the aphid leading to transmission has been well characterized, the mechanisms involved in virus recognition at aphid membranes are still poorly understood. One isolate in our collection (PLRV-14.2) has been shown to be non- or only poorly transmitted by some clones of aphids belonging...
